While Keoghan didn't give a clear yes or no answer, he gave the following cryptic response:

The Batman - Part II: News, Cast, Story, & Everything We Know
The Batman's ending teased a future film, and The Batman 2 has sure enough been confirmed. Here's everything we know about The Batman - Part IIWill Joker Be The Main Villain In The Batman 2?
While story details are being kept under wraps on The Batman - Part II, Keoghan's latest comments do raise the question of whether Joker will be the main villain in the DC movie. A case can definitely be made that The Batman did plant the seed for Joker to potentially have the role as the big bad in The Batman - Part II, following his encounter with The Riddler in Arkham Asylum. However, The Batman doesn't definitively make it clear if Joker is the next threat for Bruce Wayne to deal with in his sequel.
It is also worth taking into account that while Keoghan is now part of the franchise, having Joker as the main baddie this early could be risky for Reeves' The Batman universe. Out of Batman's villains, Joker has been depicted the most in live-action, especially in the movies, with Heath Ledger being the primary antagonist in The Dark Knight, the second movie in Christopher Nolan's trilogy. One of the reasons The Batman was such a success was because Reeves allowed a villain like The Riddler to get a proper chance to star as the first foe for the Caped Crusader, while also doing something refreshing from past Batman movies.
The Batman - Part II was supposed to start filming in November 2023, but production was delayed due to the Writers Guild of America and SAG-AFTRA strikes.
It wouldn't be shocking at all if Reeves is looking at other Batman villains for The Batman - Part II, especially those who have never been featured in a movie. From the Court of Owls to Clayface and Hush, there are so many other options to consider, as the director could always feature Joker in a larger role in a future installment. Hopefully, more news on The Batman - Part II will emerge as soon as possible to give the audience an idea of where the sequel is headed.
